Role of Notch signaling pathway in controlling parapineal size and migration during the establishment of left right asymmetry in zebrafish brain.

 

"How left right (LR) asymmetries are established during brain development is still poorly understood. In the zebrafish epithalamus, a small group of cells, called the parapineal organ (PP) arises from the anterior part of the medially located epiphysis, migrates from the midline to the left side and is required for subsequent elaboration of epithalamic asymmetries. Here, we show that Notch signaling pathway is required for both the specification and the migration of the parapineal structure. In loss of function context for Notch pathway, we observed a significant increase in the number of parapineal cells together with a defect in migration. In contrast, over-activating Notch pathway by ectopically expressing the intracellular domain of Notch receptor just before migration, causes a strong reduction in the number of parapineal cells and interferes with the migration. Our data indicate that Notch pathway tightly regulates the size of the parapineal and its migration. Our current studies aim at understanding whether Notch pathway could be required to modulate the activation of FGF pathway in the parapineal."
