I am interested in the organisation and dynamics of bacterial chromosomes and the proteins involved in this process, especially the Nucleoid-Associated-Proteins (NAPs). Using a variety of tools (microscopy, biophysics, genetics and biochemistry), I am trying to understand the process of cell division in E. coli, especially the communication between chromosome segregation and the septum closure. In particular, I am interested in the ter-linkage, with the binding of MatP in the ter region. In collaboration with C. Tardin (IPBS), I am combining a biophysical method, the Tethered Particle Motion (TPM), with short-time microscopy, which allows to generate physical properties of chromosomal loci (MSD, Diffusion Coefficient…) by analysing the motion of foci in the cell in different conditions or mutants. I am also interested in the interactions between MatP and ZapB, an element of the divisome, and their effects on chromosome segregation and advance in the process of cell division.
