Type 4 filament (Tff) nanomachines are envelope-embedded macromolecular complexes organized around a dynamic pilus-like structure. They are widespread in prokaryotes where they fulfill diverse cellular functions. One member of this superfamily, the type 2 secretion system (T2SS) promotes the specific secretion of large, folded effectors across the outer membrane through a giant, finely-controlled secretin pore, under the impetus of a small periplasmic fiber, the endopilus.
