Exposure to a high-intensity stressor can lead to the mental disorder called post-traumatic stress disorder (PTSD). In addition to hypermnesia of cue reminiscent of psychotrauma (symptom of intrusion), sleep alterations represent the first complaints of patients suffering from PTSD. These insomnias are important to better understand because they could be a risk factor and/or relapse of the disease and do not benefit from satisfactory pharmacological treatment.
The cellular and molecular mechanisms of sleep alterations induced by intense stressor are not known and, in this context, neuropeptide S (NPS) seems a relevant line of research. This presentation will highlight the unique pharmacological, anatomical and electrophysiological properties of NPS. Through an indirect synaptic action ultimately targeting the GABAergic neurons of the amygdala and the preoptic area, the administration of NPS can both facilitate the extinction processes of aversive conditioning and reduce slow-wave sleep. In perspective of this presentation, preliminary data suggesting an overexpression of the NPS system in the preoptic area (central brain area of sleep regulation) in stressed animals will be presented.
All the data presented confirm the relevance of studying the NPS in the context of PTSD and psychotrauma associated sleep disorders.
